
How psilocybin works in the brain, named papers only
Serotonin receptors, a 2006 Hopkins paper, and the limits of what a church kitchen can claim. Named sources. No miracles.
The question people type is simple. How does psilocybin work in the brain. The honest answer is longer than a label and shorter than a textbook. Psilocybin is converted in the body to psilocin. Psilocin is active at serotonin receptors, especially a subtype researchers write as 5-HT2A. That is the mechanism most papers agree to name. Everything after that sentence is interpretation, and interpretation is where brands get sloppy.
We are a sanctuary that makes chocolate. We are not a lab. We read the lab. When we say receptor, we mean a lock on a cell that a molecule can fit. When we say default-mode network, we mean a pattern of brain activity that some researchers, including Robin Carhart-Harris in work that Michael Pollan popularized for a general reader in 2018, have measured as quieter on psilocybin. Popular is not the same as settled. Settled is rare.
If you came here from a book-review essay that braided Leary, Pollan, and Tao Lin, keep the hunger and leave the sentences. Those writers earned their pages. Our job is to say what a church is allowed to repeat without pretending we ran the scanner.
The 2006 paper people actually mean
In 2006, Roland Griffiths and colleagues at Johns Hopkins published a double-blind, placebo-controlled study in the Journal of Psychopharmacology. The title people remember in short form is that psilocybin can occasion mystical-type experiences. Pollan later called that year a hinge: Hofmann's centennial, a Supreme Court case about a church and ayahuasca, and that paper. The hinge story is useful. It is still a story about other people's work.
What the paper did not do is license a kitchen. It did not prove that a chocolate square will treat depression. It did not prove that your Tuesday night will look like their protocol. Protocol means a room, a dose, a sitter, a schedule, and a consent form. We have a kitchen and a congregation. Those are different rooms.
When someone says the science is back, ask which paper and which year. 2006 is one. There have been more since, including later Hopkins and Imperial College work on depression and on imaging. Cite the one you mean. If you cannot cite it, you are advertising.
What a receptor claim can survive
A receptor claim can survive if it stays at the level of binding and downstream signaling. Psilocin fits 5-HT2A. Downstream, neurons change their chatter. People report that the usual story of self gets quieter or stranger. That report is common enough to be interesting and messy enough to be misused.
A receptor claim dies when it becomes a promise. We will not say our bar rewires you. We will not say we measured your default-mode network. We have not. If a future paper changes the picture, we will read it and we will say so. Science moves. Wrappers that pretend otherwise are lying on a delay.
Carhart-Harris and others have offered the image of a brain that has been stuck in a groove. Psilocybin, in their framing, lubricates the groove. That is a metaphor with data behind it and a metaphor that slides into sales copy if you let it. We keep the metaphor in the journal, not on the foil.
How we use this in the sanctuary
We use the science as a fence, not as a halo. The fence says: adults only, named dose, no invented indication, no stolen sentences from people who actually ran trials. The halo would say: because a paper exists, our chocolate is medicine. We refuse the halo.

Those are two different kinds of honesty.

A reading order that will not waste your night
Read the methods section of Griffiths 2006 before you read anyone's memoir of the same year. Methods are boring on purpose. Boring is where the dose, the room, and the consent live. Memoir is where the adjectives live. We like adjectives in this journal. We do not let them drive.
Then read one imaging paper, slowly. If you cannot finish it, you are not required to have an opinion about networks. You are required not to fake one. Faking a network is how a church becomes a TED talk.
Then come back to the kitchen essay in this issue. See if the kettle still makes sense after the papers. If it does, you understand our science policy. If it does not, write sanctuary@mail.herochurch.org and tell us which paper broke the kettle. We will read it. We might change a sentence. We will not change a batch to chase a headline.
Headlines expire. Receptors do not care about our marketing calendar. If you want a church that updates its theology every time a preprint lands, we will disappoint you. If you want a church that updates a sentence when a method is actually overturned, that is the pace we can defend. Pace is a scientific virtue even when you do not own a scanner.
